Intergenerational Effect of Early Life Exposure to Permethrin: Changes in Global DNA Methylation and in Nurr1 Gene Expression

چکیده
Environmental exposure to pesticides during the early stages of development represents an important risk factor for the onset of neurodegenerative diseases in adult age. Neonatal exposure to Permethrin (PERM), a member of the family of synthetic pyrethroids, can induce a Parkinson-like disease and cause some alterations in striatum of rats, involving both genetic and epigenetic pathways. Through gene expression analysis and global DNA methylation assessment in both PERM-treated parents and their untreated offspring, we investigated on the prospective intergenerational effect of this pesticide. Thirty-three percent of progeny presents the same Nurr1 alteration as rats exposed to permethrin in early life. A decrease in global genome-wide DNA methylation was measured in mothers exposed in early life to permethrin as well as in their offspring, whereas untreated rats have a hypermethylated genomic DNA. Further studies are however needed to elucidate the molecular mechanisms, but, despite this, an intergenerational PERM-induced damage on progenies has been identified for the first time.

Effects of Major Epigenetic Factors on Systemic Lupus Erythematosus
The pathogenesis of systemic lupus erythematosus (SLE) is influenced by both genetic factors and epigenetic modifications; the latter is a result of exposure to various environmental factors. Epigenetic modifications affect gene expression and alter cellular functions without modifying the genomic sequences. CpG-DNA methylation, histone modifications, and miRNAs are the main epigenetic factors ...
متن کاملEffect of Exposure to 900 MHz GSM Mobile Phone Radiofrequency Radiation on Estrogen Receptor Methylation Status in Colon Cells of Male Sprague Dawley Rats
Background: Over the past several years, the rapidly increasing use of mobile phones has raised global concerns about the biological effects of exposure to radiofrequency (RF) radiation. Numerous studies have shown that exposure to electromagnetic fields (EMFs) can be associated with effects on the nervous, endocrine, immune, cardiovascular, hematopoietic and ocular systems. In spite of genetic...

s:1993:"Background: There are many approaches that gene expression is controlled in eukaryotes. DNA methylation is one of several epigenetic mechanisms that cells use to control gene expression and lock genes in the "off" position. In addition, sperm DNA damage can correlate with DNA methylation defect. There is evidence that sperm of infertile men contain more DNA damage than fertile men and t...

Background: Because of the protection of surplus embryos, Cryopreservation is usually used in ART. It is not clear, the vitrified-warmed embryos that have normal morphology, be normal in genetic level, too. DNA methylation of gene regulatory regions can causes inhibition of gene expression. We study effect of vitrification method of cryopreservation on DNA methylation and gene expression level ...

Background: Embryo vitrification was effectively used for assisted reproductive techniques. Despite the undeniable benefits of vitrification, cooling and warming stress, and cytotoxicity of cryoprotectant may affect the DNA methylation that have an important role in gene activation and silencing. In the present study effects of 2-cell embryo vitrification on DNA methylation in hatched blastocys...
